The tumor microenvironment (TME) establishes the milieu surrounding and supporting cancer cells, incorporating various non-malignant elements such as stromal cells, signaling molecules, and blood vessels that together influence tumor evolution and treatment outcomes. Recent research is continuously revealing the complex interplay between cancer cells and the TME, demonstrating how this interaction influences cancer progression, metastasis, and therapeutic resistance. Critical elements within the TME, including the immune regulatory components, extracellular matrix, and cellular communication networks, challenge the efficacy of traditional cancer therapies by altering drug distribution and action—underscoring a significant gap in our detailed understanding of TME dynamics.

In light of these complexities, this Research Topic aims to delve deeper into the mechanisms through which therapies remodel TME and how these alterations in turn affect tumor behavior and treatment responses. Special emphasis is placed on understanding the biophysical, biochemical, and molecular changes within the TME induced by various cancer treatments—be it chemotherapy, immunotherapy, targeted therapy, or novel modalities like hyperthermia. Through this approach, the goal is to uncover novel insights that can drive the development of therapies that not only target tumor cells but strategically manipulate the TME to enhance treatment efficacy.

To gather further insights in this arena, we welcome contributions that expand our knowledge of how cancer treatments reshape the TPE and how these changes can be leveraged for therapeutic benefit. Submissions may include, but are not limited to, the following topics:
o Biochemical and molecular changes in the TME pre- and post-treatment, and their mechanisms.
o The impact of various treatment modalities on TME and their underlying mechanisms.
o Examination of key TME components and their response mechanisms to treatment.
o The influence of TME remodeling on the efficacy of cancer therapies.
o Insights into the TME's role in predicting treatment responses and patient prognoses.
o Explorations of cell and non-cell autonomous pathways affecting tumor-stromal interactions within the TME.

Please note: studies consisting solely of bioinformatic investigation of publicly available genomic/transcriptomic/proteomic data do not fall within the scope of the section unless they are expanded and provide significant biological or mechanistic insight into the process being studied and will not be accepted as part of this Research Topic.
