Chronic inflammation is a fundamental process implicated in the pathogenesis and progression of various complex immune diseases, often resulting in adverse outcomes like fibrosis. Current immunomodulatory therapeutics have made significant steps forward in treating such inflammatory conditions, yet they often hit an efficacy ceiling. This reflects gaps in our understanding of the heterogeneous molecular pathogenic drivers underlying these diseases. A more granular classification of patients based on inflammation-related molecular signatures can pave the way for precision medicine, enabling the tailoring of treatments to individual molecular phenotypes. Furthermore, insights gained through advanced data analytic methods can accelerate the development of next-generation therapies.

This Research Topic focuses on the urgent need to advance our understanding of inflammation's role in immune diseases and to apply these insights to precision medicine. The aim is to address the limitations of current model systems and directly study human immunopathologies, utilizing the power of patient-derived data and advanced analytical technologies. By identifying inflammation-driven molecular signatures and resistance pathways, this work will help improve therapeutic outcomes and guide drug development strategies.

1. Deconvolution of Inflammatory Drivers in Immunological Diseases
a. Identify mechanism-based biomarkers associated with inflammation and disease progression.
b. Clarify how inflammatory pathways contribute to disease dynamics and therapeutic resistance.

2. Identification of Prognostic Inflammatory Biomarkers
a. Develop biomarkers that predict future risk and progression of inflammatory immune diseases.
b. Enhance patient stratification and selection criteria for clinically relevant inflammation-focused studies.

3. Reverse Translational Approaches
a. Implement bedside-to-bench methodologies focusing on inflammation to reveal mechanisms behind resistance to current therapies.
b. Investigate and characterize inflammation pathways in dermatology, gastroenterology, and rheumatology, providing insights for precision medicine.

This Research Topic seeks to drive forward our understanding and treatment of immunological diseases, emphasizing clinically meaningful outcomes like therapeutic responses and patient stratification.
