Endocrine and metabolic disorders, including conditions like obesity, diabetes, hyperlipidemia, metabolic dysfunction-associated steatotic liver disease, and adrenal disorders, are a significant public health burden affecting a large swath of the population. Medications remain the linchpin in the management of these complex disorders but come with their own sets of challenges, such as variable patient response, drug interactions, and side effects. A particularly pressing concern is that many medications themselves can cause damage to endocrine glands, thereby exacerbating the existing hormonal imbalances or creating new ones.

For example, corticosteroids, often used to treat inflammatory conditions, can lead to adrenal insufficiency or Cushing's syndrome. Likewise, some antipsychotic medications are associated with hyperprolactinemia or even insulin resistance. Understanding the mechanisms and scope of these medication-induced endocrine damages is crucial for effective treatment. This adds another layer of complexity and urgency to the push for personalized treatments of endocrine disorders.

Innovations in pharmacogenomics, bioinformatics, and patient stratification offer promise for individualized treatment plans that minimize side effects while maximizing efficacy. However, these advanced techniques are still not integrated adequately into mainstream clinical practice. Moreover, scant research is currently directed towards personalized management strategies specifically, aiming at countering the endocrine-related side effects of medications.

The primary objective of this Research Topic is to address the critical gaps in our understanding of medication-induced damage to endocrine glands and the broader physiological system. Concurrently, we aim to fast-track the implementation of personalized treatment regimens that factor in pharmacogenomics and patient stratification. By utilizing an interdisciplinary approach and leveraging recent advances in advanced algorithms and cross-disciplinary research, the goal is to create evidence-based guidelines for personalized treatments. These strategies should serve dual purposes: effectively manage the primary endocrine disorder and minimize medication-induced endocrine side effects, thus offering a more tailored and holistic approach to patient care.

This Research Topic aims to cover a comprehensive study of endocrine diseases, with an emphasis on the adverse impacts of medication on endocrine glands and the advancement of personalized treatment strategies. We invite authors to submit original research, comprehensive reviews, methodological papers, and clinical case reports focusing on the following themes:
• Mechanisms of Medication-Induced Endocrine Damage: Research should clarify the cellular and molecular pathways through which drugs disrupt endocrine function.
• Pharmacogenomics and Personalized Treatment: Studies should focus on the influence of genetic information on drug responses and the development of personalized treatment plans based on this data.
• Patient Stratification: Papers should explore risk factors, biomarkers, or other criteria that could classify patients into different treatment subgroups.
• Application of Advanced Algorithms: Submissions should discuss how advanced algorithms can decode complex clinical data to inform more personalized treatment strategies.
• Interdisciplinary Research: Collaborative studies involving endocrinology, pharmacology, genetics, and data science are encouraged.
• Real-world Applications: Manuscripts should detail the clinical outcomes or health economics of implementing personalized treatments for endocrine disorders.
