Cancer is the second leading cause of death worldwide, and the incidence of cancer is increasing every year. Lipids, including fatty acids, cholesterol, and phospholipids, play a major role in cancer progression and metastasis, as they are involved in energy production, signaling pathways, and biogenesis of cell membranes. The synthesis, storage, degradation, and uptake of lipids are dysregulated in cancer, leading to tumour cell proliferation, survival, immunogenicity, and metastasis. Altered lipid metabolism impairs membrane composition, signaling pathway activity, gene expression, energy metabolism, oncogenic pathway activation, and cell death pathways in various cancers, including prostate, lung, ovarian, colon, gastrointestinal, breast, and brain cancers. Targeting lipid metabolic pathways, lipid metabolism-related enzymes, and molecular mechanisms involved in lipid metabolism has shown promising anticancer effects. Revealing the possible mechanisms behind lipid metabolism rewiring in cancer is crucial to develop therapeutic strategies in the future.

Since altered lipid metabolism promotes cancer initiation, progression, and metastasis, developing novel therapeutic approaches to target lipid metabolism can be a promising approach to treat cancer. In this research topic, we aim to improve our comprehensive understanding of the regulatory role of lipid metabolism in cancer cell proliferation, survival, metastasis, immunogenicity, and tumour microenvironment. This may provide a new direction and novel perspectives for further studies to develop promising therapeutic approaches and strategies for various types of cancers. Current studies aim to develop novel anticancer strategies distinct from traditional approaches to prevent and treat cancer worldwide. In this special topic, researchers are encouraged to submit original research articles to improve our knowledge about the role of lipid metabolism in cancer progression and metastasis with novel perspectives. Original research and review articles describing the state of the art and systematic reviews on this topic are also encourage.
