Innate immunity has been extensively recognized and acknowledged as an important coordinator for immunosurveillance and antitumor immunity. There is growing evidence that a diverse cadre of innate immune cells and molecules participate in regulating proinflammatory programs and bridging adaptive immunity, thereby determining the outcome of cancer immunosurveillance and patient survival. For instance, the status of myeloid-derived cells plays an important immunoregulatory role in anti-tumor immunity. Dendritic cells are essential for adaptive immune responses via potentiating CD8+ T cells with tumor-associated antigens and costimulatory molecules as well as CD4+ T cell recruitment in the tumor. Tumor-associated macrophages and neutrophils can exert antitumorigenic or protumorigenic effects, depending on their polarization to proinflammatory type 1 or anti-inflammatory type 2 cells. Natural killer cells actively perform direct cytotoxic functions independent of antigen-specific recognition of the tumor. In these cellular processes, many factors have been identified as regulators, such as pattern recognition receptors (e.g. TLR7/8/9, RIG-I, MDA-5, STING, NLRP3, etc.), cytokines (e.g. IFNγ, TGF-β, etc.), chemokines (e.g. CSF-1R, etc.) and immune regulators (e.g. CD40, CD47, CD209, PI3K, etc.). Many novel therapeutic strategies have been investigated by targeting these key innate immunity processes. Nevertheless, the precise regulatory cellular and molecular mechanisms of tumor associated innate immunity remains largely under investigation.
The goal of this Research Topic is to provide a forum to discuss the molecular intersection between innate immune regulators and oncogenic pathways, thus providing insights into the mechanisms of proinflammatory programs orchestrated by innate immunity, the crosstalk between innate and adaptive immune responses as well as the development of new therapeutic strategies targeting innate immune regulators.
