PTEN deletion in postnatally generated granule cells in the dentate gyrus of the mouse hippocampus is sufficient to cause seizures (Pun et. al, 2012, Neuron). Video shows a three dimensional rotational image of fluorescently labeled cells in which PTEN has been deleted from the edge of the upper blade of the dentate gyrus. Close up view shows anatomical characteristics of these mutated cells, notably the presence of basal dendrites and soma hypertrophy. Mossy fibers (axons of granule cells) and mossy fiber boutons are clearly visible in the image. Images were taken using a Nikon A1R MP upright confocal microscope in both single and multiphoton modes with an Apo LWD 25x water immesion objective. Image was depth coded using ImageJ software which produces the spectrum of colors. Image depth is 500um.
