Background

The molecular master modulators and mechanisms that regulate metabolism include nitric oxide (NO) and superoxide anion radicals (SOR). Recent studies examining L-arginine and NO, have demonstrated anti-ageing effects by improving blood flow, maintaining endothelial health and stimulating mitochondrial activity and biogenesis.

Ageing is often associated with increased adiposity, altered and reduced muscle mass or sarcopenia including increased ectopic fat stores, some of which are independently associated with increased cardiometabolic risk and physical dysfunction. The development of insulin resistance is a feature of endothelial dysfunction, which has been observed with age associated metabolic diseases such as obesity and sarcopenia. A reduced NO formation by insufficient supply of L-arginine leads to an endothelial dysfunction that compromises the endogenous supply of energetic resources to enable regeneration. The age dependent increase of asymmetric dimethylarginine levels and the upregulation of L-arginine depletion by enhanced arginase activity, are important factors in the alteration of the L-arginine/nitric oxide pathway. This may lead to insulin resistance and endothelial dysfunction with impaired vasodilation. Such changes are promoting degenerative processes that can lead to disorders and diseases associated with enhanced morbidity and mortality.

The current challenge in this field is to target such inflammaging successfully. New approaches will have to modify L-arginine metabolism specifically and will likely combine L-arginine with other agents such as antioxidants. The progress in the field will depend on the development of such a precision supplementation.

This Research Topic will focus on the latest developments in the integrative pharmacology of regeneration enabled by L-arginine and NO as modulators of cardiometabolic control and redox regulation; examining and reporting on the latest findings on L-arginine as a physiological and pharmacological agent.

We welcome manuscripts biochemical, experimental and clinical studies and reviews addressing the health promoting effects of L-arginine. Areas of interest include, but are not limited to, effects of L-arginine on the cardiovascular system, fat and protein metabolism, the gastrointestinal tract, immune system and liver, wound healing and neurovascular coupling and cognition.
