Background

Both photodynamic therapy (PDT) and immunotherapy are two of the biggest areas of cancer therapy with the potential to revolutionize healthcare. PDT conveys the idea of combining photosensitizer agent with light (laser) to provide a holistic solution for various malignant and non-malignant diseases management. Also, cancer immunotherapy is a type of treatment that harnesses the body's immune system to recognize and fight cancer cells by providing specific components, such as immune checkpoint inhibitors, antibodies or vaccines to better detect and attack cancer. Each of these therapies works by different mechanisms, but when combined, they have the potential to enhance treatment outcomes, especially in overcoming resistance mechanisms that limit the effectiveness of traditional therapies. However, to promote cancer therapy and overcome cancer resistance after common treatments such as radiotherapy and chemotherapy, using PDT and immunotherapy as a non-invasive strategy are critical. PDT is a cancer treatment that uses a photosensitizing agent, which is activated by a specific wavelength of light and produce reactive oxygen species (ROS) that cause damage to the tumor cells. However, despite its effectiveness, resistance to PDT can develop. Some tumor cells may repair the damage caused by ROS, while others may develop mechanisms to evade immune system activation, limiting the long-term efficacy of PDT. During immunotherapy, cancer cells can evade immune detection through various mechanisms, such as low immunogenicity, expression of immune checkpoints, or the presence of an immunosuppressive tumor microenvironment. Combining PDT with immunotherapy addresses the limitations of each individual approach and has several synergistic effects.
