Background

Bone regeneration and remodeling are imperative for the sustenance of osseous function and structural integrity. The human bone architecture is subject to incessant osteotropic activities, oscillating between osteoblastic deposition and osteoclastic resorption. For example, the alveolar bone goes through the bone remodeling process during orthodontic treatment. Moreover, bone defects attributable to pathological conditions or traumatic events necessitate intervention via osteoregenerative processes. Although osteogenic tissues possess an inherent regenerative capacity, bone deficits exceeding a critical magnitude compel the incorporation of adjunctive measures such as osteoconductive scaffolds, osteoinductive growth factors, and osteogenic cellular elements. It is quintessential to elucidate the biological mechanisms at play during osteogenesis and bone remodeling. Concurrently, the burgeoning domain of biomaterials proffers pivotal methodologies to augment these endogenous processes, thereby fostering enhanced clinical outcomes. In addition, the confluence of biomaterials with biological entities engenders synergistic strategies for the innovation of therapeutic modalities that potentiate bone remodeling and osteogenesis.

To promote the effectiveness and efficiency of bone regeneration and remodeling, it is important to better understand the biological regulation mechanism and improve the biomaterial properties. Various biomaterials have been applied in promoting bone regeneration and remodeling, such as metals, bioceramics, scaffolds, hydrogels, 3D printing, and microspheres, but their properties can be enhanced to get higher biocompatibility, less adverse reactions, and more osteoinductive to promote cellular proliferation and differentiation. Lots of extracellular and intracellular signaling pathways such as WNT, BMP, and FGFR have been studied to figure out the biological mechanisms, but it is still not fully understood. This research topic aims to present current interesting studies about bone regeneration and remodeling with the approaches of advanced novel biomaterials or various biological manipulations. Thus, providing references and support for future clinical applications.
