Gliomas are central nervous tumours with a glial cell of origin, such as neural stem cells, astrocytes and oligodendrocytes. They usually occur in the brain (supratentorial, infratentorial, brainstem) but occasionally originate in the spine. Gliomas may be benign or malignant, comprise of various histopathological subtypes and WHO grades (I-IV). Despite having a low incidence rate of around 3.19 per 100,000 (in the US) and a median age of 64 years for a first diagnosis, gliomas account for 80% of malignant brain tumours, and are most often fatal; they are the leading cause of cancer-related death among males from the ages of 0-39 and in females aged 0-19.
Though long-term studies have shown a decrease in both confirmed cases and in mortality over the last 20-30 years, it is also true that the future burden of this disease on both healthcare systems and economies may become increasingly significant. Studies on the genetics of gliomas continue to provide hope for better treatment and healthcare solutions. New gene signatures and biomarkers are still being discovered that can be used to improve all stages of the disease, from earlier diagnoses to potentially more targeted medicines and therapies. The advent of next-generation sequencing, as well as higher resolution DNA copy number and methylation profiling technologies, have revolutionized the interrogation and understanding of somatic profiles of glioma and spatial and temporal aspects to the genomic heterogeneity of gliomas. Genetic profiling of glioma has led to several proposed subtypes beyond the histopathological grading system and the WHO recognizes genetic screening as an integral part of clinical decision making and diagnosis of glioma based on both genetic and morphologic factors.
This Research Topic will aim to attract high quality manuscripts based around the topic of the genomics and molecular genetics of glioma. The collection will aim to serve as a hub of knowledge for clinicians and researchers, where insights and key advances in genomics coupled with other clinical information such as histopathology, radiographic imagining, and patient chart information can be presented and shared, and progress can be made towards the eventual goal of a complete understanding of this complex disease.
We propose a collection of articles on the following aspects of glioma genetics:
• Characterization of genetic factors in glioma pertaining to immunity and immune-related factors such as immunotherapy and immune microenvironment.
• Experimental discovery and validation of pan-target, multi-gene signatures in glioma as prognostic and predictive markers.
• Experimental discovery and validation of single-target, single-gene characterization in glioma prognostic and predictive markers.
Please note: manuscripts consisting solely of bioinformatics or computational analysis of public genomic or transcriptomic databases which are not accompanied by validation (independent cohort or biological validation in vitro or in vivo) are out of scope for this section and will not be accepted as part of this Research Topic.
