As a therapeutic strategy, cancer immunotherapy is now increasingly popular, treating cancer through the utilization of the human immune system. These regimens include tumor vacancy, recombinant immune cytokines, adoptive T cell transplantation (T cell, TCRT, CART, etc.), as well as immune checkpoint blockade. The efficacy of cancer immunotherapy, however, was largely limited because tumor cells may develop multiple approaches to evade immune surveillance by hijacking the hosts immune system. In recent years, immune checkpoint blockade therapy (ICBT), which blocks tumor immune escape, has exhibited momentous clinical benefits, placing tumor immunotherapy under the spotlight. Several immune checkpoints have been discovered to be optimal targets for immune blockade, including the cytotoxic T-lymphocyte-associated protein 4 (CTLA-4) and programmed cell-death protein 1 (PD-1)/programmed cell-death 1 ligand 1 (PD-L1) pathways. Nevertheless, not only the limited tumor response, usually between 5% to 40%, but also acquired drug resistance have been observed in patients receiving long term ICBT treatment, highlighting an urgent need to extend our understanding on the mechanisms that regulate anti-cancer immunity.
