The NLR family pyrin domain containing 3 (NLRP3) inflammasome is an intracellular multiprotein complex that has a vital role in activating and regulating innate immune responses. The activation of NLRP3 can induce the production and release of pro-inflammatory IL-1b and IL-18 cytokines accompanied by an inflammatory cell death known as pyroptosis. The tight regulation of NLRP3 inflammasome activity is essential in the host defense against invading microbes, sterile danger signals, and environmental perturbations. The immunopathogenesis of several metabolic, neurodegenerative, and autoimmune diseases has been linked to dysregulation in the NLRP3 inflammasome.
This Research Topic aims to provide an update on the mechanisms underlying the NLRP3 inflammasome activation of the innate immune system in various pathologies. We will also be interested in submissions focusing on the activation of the NLRP3 inflammasome by soluble immune mediators, and therefore, the regulation of NLRP3 and innate immune responses. Understanding the mechanisms of action of NLRP3 will be important in developing novel NLRP3 targeted inhibitors and therapies to advance the treatment of disorders caused by abnormal inflammasome activation.
In this Research Topic, we welcome the submission of Original Research, Review, and Mini-Review articles focusing on NLRP3 Activation in Innate Immune Responses. We welcome submissions covering, but not limited to, the following sub-topics:
• Novel molecular and mechanistic insights into the role of NLRP3 in activating innate immune responses
• The role of soluble innate immune mediators in NLRP3 activation
• NLRP3 regulation of innate immune responses
• Novel therapies targeting NLRP3 activation/regulating in controlling pathological innate immune responses
• The use of cutting-edge technologies in improving our understanding of the role of the NLRP3 in activating/regulating innate immune responses
