Our knowledge of the modulation of brain homeostasis is rapidly increasing. Perturbations of this delicate equilibrium result in alterations of the nervous system, affecting a variety of fundamental activities: learning and memory, cognition, intellectual capacities, emotions but also peripheral functions modulated by the nervous system, such as motor activity and gastrointestinal efficiency. A deep understanding of the mechanisms involved is needed for treating and possibly preventing these negative consequences. New links between brain health and general wellness are emerging. Not only the list of toxic compounds is expanding, but other parameters, including lifestyle and environmental frameworks, are increasingly pointed as key factors. Considering intellectual and physical activity, food intake, drug consumption (including new synthetic drugs), and exposure to multiple contaminants is becoming a necessity to better understand brain health. Inflammation, oxidative stress, excitotoxicity, and mitochondrial stress are some of the processes involved in brain disease and that contribute to the appearance of pathologies such as Alzheimer’s disease and dementia, epilepsy, SLA, Parkinson, and many others.
This Frontiers Research Topic proposes to collect articles that are elucidating mechanisms of neurotoxicity, as well as epigenetic modulation of key regulatory pathways but also emerging parameters that can influence different risk settings such as microbiome composition. Particular attention will be given to studies focusing on innovative approaches evaluating known toxic agents or emerging ones.
This Research Topic is open for original research articles using different approaches to evaluate the impact of new and emerging factors influencing brain health and the underpinning mechanisms. We also encourage submissions of theoretical approaches such as Reviews, Perspective, and Opinion articles on promising future directions in the field.
We aim to collect manuscripts on the following topics:

- impact of drugs on brain health and disease
