Identification and functional characterization of the MYB transcription factor GmMYBLJ in soybean leaf senescence

Leaf senescence is an important agronomic trait that significantly influences the 两端对齐 设置格式[Bsm]: 删除[Bsm]: analysis 删除[Bsm]: analysis 删除[Cui, Yuhai (AAFC/AAC)]: characterization 删除[Bsm]: associated with leaf senescence 删除[Cui, Yuhai (AAFC/AAC)]: in soybean 删除[Bsm]: Unclassified / Non classifié quality and yield of soybean. v-Myb avian myeloblastosis viral oncogene homolog (MYB) transcription factors are considered as crucial regulators governing leaf senescence, which can be utilized to improve agronomic traits in crops. However, our knowledge regarding the functional roles of soybean MYBs in leaf senescence is extremely limited. In this study, GmMYBLJ, a CCA1-like MYB, was identified and functionally characterized in leaf senescence. The GmMYBLJ protein is localized in the nucleus, and high accumulation of its transcriptswas observed in nodules and embryos. Noticeably, GmMYBLJ was highly expressed in soybean senescent leaves and transcriptionally induced by dark or NaCl treatment, which was confirmed by histochemical GUS staining analysis. Ectopic over-expression of GmMYBLJ in Arabidopsis not only led to earlier leaf senescence, less chlorophyll content and more MDA accumulation, but also promoted the expression of several several WRKY family transcription factors andsenescence-associated genes such as SAG12 and ORE1. Further investigation showed that over-expression of GmMYBLJ accelerated Arabidopsis leaf senescence under darkness and in responding to Pst DC3000 infection. Moreover, transgenic soybean plants over-expressing GmMYBLJ grew faster and exhibited accelerated senescence under salt stress. DAB staining analysis showed that GmMYBLJ conferred ROS accumulation in soybean hairy roots and Arabidopsis leaves. Collectively, our results provided useful information on the functional roles of GmMYBLJ in both aging-dependent and stress-induced senescence.
