Establishment of Regeneration System of Peucedanum praeruptorum and Molecular Association Analysis of Coumarin Secondary Metabolism

    The objective of this study is to ascertain the optimal conditions for the regeneration system of Peucedanum praeruptorum, and to establish the relationship between the distribution of coumarin content and the expression of key genes. Firstly, this work investigated the effects of the type, concentration, and proportion of exogenous biological hormones on the induction, proliferation, differentiation and rooting of P. praeruptorum. We demonstrate that MS + 2,4-D 0.5 mg/L + 6-BA 0.5 mg/L was the optimal induction medium for callus, MS + 6-BA 1.0 mg/L + IBA 0.5 mg/L was the optimal proliferation medium for callus and MS + 6-BA 0.5 mg/L + IBA 0.5 mg/L was the optimal differentiation medium for adventitious buds. MS + IBA 0.5 mg/L was the most suitable rooting medium for tissue culture seedlings and the survival rate was higher when using the MS liquid medium with the filter paper bridge method. Furthermore, on the basis of the successfully established regeneration system for P. praeruptorum, an analysis was conducted of the relationship between the distribution of its active components of coumarin and the expression of key genes.
