Multiple studies suggest that impaired 1,25(OH) 2D3-VDR signaling may play a key role in the development and progression of thyroid cancer. Integrated analysis mRNA expression infor-mation from The Cancer Genome Atlas and GEO, we compared gene expression in cancer and normal tissues and identified differentially expressed genes (DEGs). We calculated risk estimates for the seven genetic markers and constructed predictive models using LASSO-Cox regression tested the predictive value of the genetic markers. There were 64 calcium metabolism-related genes showing differences between tumor tissue and normal tissue. Ten cases of GDE were sig-nificantly associated with overall survival. Patients were divided into high- and low-risk groups based on the average risk score for seven genetic markers. Low-risk patients had significantly better overall survival than high-risk patients. Cox regression analysis showed that risk levels had independent predictive power. Functional analysis showed that the immune status of the two groups was different. Moreover, we also investigate the effect on thyroid cancer biological func-tions, immune microenvironment and drug resistance of these calcium metabolism-related genes. Our data predicted a novel genetic signature associated with calcium metabolism predicts prognosis in patients with PTC.
