FBXO22-mediated ubiquitination of multiple proteins has been reported in tumorigenesis. However, it is unclear whether FBXO22 plays a critical role in the pathogenesis of different cancers through certain common molecular mechanisms. Furthermore, there is a lack of clinical evidence for the association between FBXO22 and pan-cancer. In this study, we discovered that the aberrant expression of FBXO22 in human cancers is associated with a poor clinical prognosis. To explore the regulatory role of FBXO22 in tumorigenesis, we first collected 23 FBXO22-binding proteins through the STRING database and literature search. These genes are significantly enriched in cell cycle negative regulation and ubiquitination pathways. Overexpression of FBXO22 enhanced the protein level of cyclin-dependent kinase 4 (CDK4) and promoted the proliferation of lung cancer cells A549. In addition, we observed an enhanced expression level and a reduced methylation level of FBXO22 in multiple tumors. High expression or low methylation of FBXO22 is associated with both increased risk and low TILs abundance in patients with multiple cancers. In conclusion, our findings have important implications for exploring the pathogenesis or clinical prognosis of FBXO22 in different tumors.
