The cytotoxicity of most chemotherapeutics or targeted drugs in the treatment of osteosarcoma is partially attenuated. And there is a poor response to current chemo- and radiotherapy at both primary lesions and pulmonary metastases of osteosarcoma. Thus,it is extremely necessary to explore promising candidates which could improve the efficacy of anti-osteosarcoma. Autophagy, a dynamic and highly conserved catabolic process, is a double-edged sword, both promoting cell survival and cell killing. Currently, the dual role of autophagy has been wider reported in different tumors, and more and more researches pointed out the potential valuable for using autophagy in clinical therapy. Here we provide a discussion of autophagy in osteosarcoma with regard to the modulations of autophagy, the role autophagy played as well as the significance of targeted autophagy in osteosarcoma therapy.
