Migrasome Regulator TSPAN4 Shapes the Suppressive Tumor Immune Microenvironment in Pan-Cancer

Migrasomes are newly discovered organelles located on the retracting fibers of migrating cells. They release various signaling molecules into the environment, expel damaged mitochondria from the cell, or are phagocytosed by receptor cells to complete intercellular information transmission. TSPAN4 serves as a highly valuable marker for visualizing migrasomes and is also an important regulator of migrasome formation. Currently, the function of TSPAN4 in cancer remains unclear. Here, we have discovered that TSPAN4 is aberrantly expressed in various tumors and may be closely associated with its methylation level. Additionally, we have revealed its prognostic significance in pan-cancer and identified its strong correlation with tumor heterogeneity and stemness, along with its involvement in shaping the suppressive tumor immune microenvironment through analysis of RNA-seq and scRNA-seq data.Furthermore, we conducted a detailed investigation of TSPAN4's involvement in glioma and determined that it affects glioma cell proliferation and facilitates the polarization of macrophages toward the immunosuppressive M2 type.
