However, ILC subsets infiltrating in ischemic myocardium, the roles of ILC subsets playing in myocardial infarction (MI) and myocardial ischemia-reperfusion injury (MIRI), and the related cellular and molecular mechanisms, have been described far from being desired in detail. Method: In the current study, the myocardia of normal, MI and MIRI mice were studied at a single-cell resolution. Flow cytometry was used to identify the existence of new ILCs clusters. Results: Five ILC subsets were found, including ILC1, ILC2a, ILC2b, ILCdc and ILCt. It’s worth noting that ILCdc, ILC2b and ILCt were identified as new ILC subclusters. The cellular landscapes of ILCs were revealed and signal pathways were predicted. What’s more, pseudotime trajectory analysis exhibited different ILCs statuses and traced related gene expression in normal and ischemia conditions. In addition, we established ligands-receptors-transcription factors-target genes regulatory network to disclose cell communications among ILCs clusters. Moreover, we further illuminated the transcriptional features of ILCdc and ILC2a subsets. Finally, the existence of ILCdc was identified by flow cytometry. Conclusion: Collectively, by mapping the spectrums of ILC subclusters, our results provided a new blueprint for understanding ILC subclusters’ roles in myocardial ischemia diseases and further potential treatment targets.
