ABSTRACT=Acute lung injury (ALI) and acute respiratory distress syndrome (ARDS) are a continuum of lung injuries attributed to various reasons. Recently, growing evidence has associated mitochondrial dysfunction with ALI development; in particular, mitochondrial DNA(mtDNA) released from impaired mitochondria plays a crucial role in the inflammatory response and histopathological changes in the lung. In this review, we evaluate the pathogenic mechanisms of mtDNA in innate immune signaling pathways and possible therapeutic targets in ALI/ARDS.
