MicroRNA-21-5p profiles in the alveolar bone of medication-related osteonecrosis of the jaw following tooth extraction in a rat model

    Objective This study aimed to compare microRNA-21-5p expressions at the extraction wound of the maxillary bone between medication-related osteonecrosis of the jaw (MRONJ) rats and normal rats at different time points.Materials and Methods 18 female, 8-week-old Sprague-Dawley rats were randomly assigned to experimental group (n=9) and control group (n=9). To establish a MRONJ rat model in maxillary right first molar area in experimental group, zoledronate (66 µg/kg) plus dexamethasone (5 mg/kg) were given intraperitoneally every other day for 2 weeks before tooth extraction. Normal saline was administered in a control group. After tooth extraction, drugs were continuously given until experimental endpoints which are 1-, 14-, and 28-days post tooth extraction. At each end point, 3 rats from each group were euthanized. The maxilla bones at the wound area were harvested. Real-time polymerase chain reaction (RT-PCR) was performed to compare the expression levels of miRNA-21-5p at each time point between the MRONJ group and normal group.Form gross appearance, rat received zoledronate and dexamethasone developed MRONJ as shown by non-healing wound and exposed bone at 14and 28-day post extraction in contrary to the controls. RT-PCR showed expression level of miRNA-21-5p was relatively higher in MRONJ rats compared to controls rats at day 14 and then the difference was lowered at day 28 post tooth extraction.The findings indicated that microRNA-21-5p expression levels varied during the socket healing process in MRONJ rats, reaching a peak at two weeks after tooth extraction.
