Amygdaloid acylated-ghrelin injections induce changes in serum metabolit concentrations

Acylated-ghrelin (A-Ghr) is a brain-gut peptide involved in the regulation of feeding. In our previous experiments it was verified that the intraamygdaloid application of A-Ghr dose dependently decreased liquid food intake. In the present experiments the possible effects of intraamygdaloid A-Ghr on blood plasma concentration and other feeding related metabolites were examined. In ad lib. fed male Wistar rats the effects of bilateral intraamygdalar application of 50 or100 ng A-Ghr on blood glucose level (Glucometer Elite 2000, Bayer) were measured 10 min before and 10, 20, 30, 50, 70, 90, 120 min after the microinjections. A-Ghr was dissolved in sterile saline and applied in 0.4 µl. Fifty ng or 100 ng A-Ghr treatment caused significant elevation of blood glucose concentration at the 20th and 30th or at the 10th and at the 20th min, respectively, compared to controls. In separate experiment we also measured the effect of A-Ghr on serum total-cholesterol, HDL, triglyceride, insulin and leptin concentrations, 20 min after bilateral intraamygdaloid injections of 50, 100 ng A-Ghr or saline. Significant decrease of total-cholesterol and HDL were observed after the administration of both doses of A-Ghr compared to control treatment. In addition the application of 50 ng A-Ghr decreased the serum insulin concentration. There was no any alteration in the serum level of triglyceride or leptin. Our data are the first reporting that the intracerebral injection of A-Ghr initiates changes in the peripheral concentration of metabolites.
