Obesity and inflammation. The effect of Resveratrol.

Obesity is one of the most important pandemics of the XXIst century, featured by the unbalanced control between food intake and energy consumption. Manifestations of interest involving this pathology include the release of reactive oxygen species (ROS) and inflammatory factors such as TNF-α and IL-6 that contribute in the development of insulin resistance and type II diabetes. The purpose of our research is to evaluate the anti-inflammatory effects of Resveratrol in the differentiation of pre-adipocytes in 3T3-L1 mice.
The methodology consisted in the incubation of 3T3-L1 mice pre-adipocytes in the presence of a differentiation cocktail that included Rosiglitazone, Isobutylxanthine and Dexamethasone. Observations were made every two days in a period of time between 0 and 10 days. Doses of Rosiglitazone (1μm) and Resveratrol (50μm) were applied afterwards and new observations were made until the 6th day to corroborate the effect of the Resveratrol.
The presence of triglycerides was determined by both qualitative and quantitative tests. The qualitative test was done with the use of red oil while the quantitative test consisted in the extraction of lipids with 5mL of isopropanol to then read the rate of absorption of the extracted liquid at 510nm in an Eppendorf biophotometer. Once the results were obtained, the Anova test was applied to elaborate the statistics analysis.
The obtained results, both qualitative and quantitative, demonstrated that cells treated with Resveratrol contained fewer levels of triglycerides than cells where Rosiglitazone was administered. This is due to phytoalexin, a substance that stimulates the action of the Sirtuin-1 enzyme (Sirt-1), that inhibits the nuclear receptor PPARg, avoiding the process of adipogenesis and the production of inflammatory factors, making this technique a potential therapeutic alternative.
