Application of electrophoresis and western blot to detect IgG in CSF of patients with multiple sclerosis

Multiple sclerosis (MS) is a demyelinating autoimmune disease of the central nervous system, characterized by the spread of multiple lesions in the white matter. Its diagnosis is mainly based on clinical and radiological signs, but laboratory tests based on analysis of IgG in cerebrospinal fluid (CSF) and serum are required for diagnosis with more certainty. The purpose of this study is to test the feasibility of adopting the technique of electrophoresis (PAGE: polyacrylamide gel electrophoresis) followed by western blot in the diagnosis of this disease (MS). For this we collected the couples serum / CSF of a population composed of 10 patients aged from 18 to 56 years with clinical and radiological suggestive of MS and controls (n = 3) with other non-inflammatory neurological diseases. The proteins of the two fluids (serum and CSF) are extracted and separated by various variants of PAGE followed by immunoblotting. The comparison between the protein patterns of CSF of patients and those of controls suspects the presence of IgG in the CSF of patients within the zone of high molecular weight on SDS-PAGE (dodecyl sulphate- gel electrophoresis). Using the technique of Western blotting confirmed the presence of IgG in the CSF of patients. An epidemiological study has determined the sex ratio (1: 2.3), the average age (35.68) and the most common form (relapsing-remitting) of this disease.
