IMPAIRMENT OF MEMORY FORMATION, CONSOLIDATION AND RETRIEVAL OF INHIBITORY AVOIDANCE TASK IN ACUTE MITRAGYNINE-TREATED RATS

Mitragynine, the major alkaloid compounds of Mitragyna speciosa Korth from Rubiaceae family has been widely used throughout Southeast Asian countries to enhance tolerance for working under scorching sun, to reduce pain sensation and as an alternative for treating morphine addicts. However, the availability of scientific information regarding the cognitive functions is scarce. Hence, the present study aims to investigate the effects of acute mitragynine administration on different stages of learning and memory processes called memory formation, consolidation and retrieval in inhibitory avoidance task. Male Sprague-Dawley rats (180-250 g) were given pre-training, post-training or pre-test intraperitoneal administration of mitragynine at different doses (1, 5 and 10 mg/kg) and tested for passive avoidance task (footshock: 0.5mA, 10 s). Step-through latency (s) was taken as a measure of retention score. The results showed similarity in memory formation, consolidation and retrieval process. Mitragynine significantly reduces the step-through latency at all doses in comparison with vehicle group, but there is no difference as compared to morphine-treated group (5 mg/kg, ip). No significant difference in retention score was also observed between 1, 5 and 10 mg/kg of mitragynine-treated groups. Results obtained suggest that acute administration of mitragynine does impair the memory formation, consolidation and retrieval of inhibitory avoidance task in rats.
