Synergistic effect of Centella asiatica (L.) and neurotrophic factors on neural differentiation of in vitro human mesenchymal stem cells (hMSCs)

Centella asiatica (L.), is known as a nerve tonic since ancient time, however, till date there are very few scientific evidences on this claim. Our objective is to determine that Centella asiatica (L.) can promote neural differentiation of hMSCs. In this study, Wharton’s jelly derived-hMSCs were cultured and differentiated to Schwann-likes cells using different concentrations of bioactive extract of Centella asiatica (L.) (BECA) with or without the neurotrophic factors. Cell morphology was observed under inverted microscopy, while presence of neural markers; namely GFAP, p75 NGFr, MBP and S100β were determined by immunocytochemistry analysis and quantitative polymerase chain reaction (qPCR). The results showed that neural differentiation of hMSCs was more distinct in hMSCs induced with combination of BECA and neurotrophic factors compared to BECA or neurotrophic factors alone. It seems that BECA reacts synergistically with the neurotropic factors in promoting neural differentiation of hMSCs. This finding suggests that BECA could be an effective accelerant to the neurotrophic factors in facilitating neural regeneration.
