Possible involvement of alterations in tumor necrosis factor beta secretions in the metformin-induced suppression of the proliferation in P19 cells

Neural stem/progenitor cells (NPCs), which are defined by their ability to self-renew and differentiate into 3 major cell types including neurons, astrocytes, and oligodendrocytes, play a key role in the development and maturation of the central nervous system. P19 embryonic carcinoma cells resemble NPCs. It has been reported that 5’-AMP-activated protein kinase (AMPK) acts not only as an efficient sensor for cellular ATP level, but as a regulator for cellular proliferation and differentiation. Here, we measured cytokine expressions in P19 cells under the condition of the metformin-induced suppression of the proliferation. P19 cells were cultured in alpha minimum essential medium in a 5% CO2 humidified chamber at 37°C. After plating P19 cells at a concentration of 3.0×105 cells/mL, at every 12 h, cell viability was measured by abilities of MTT reduction and gene expression was semi-quantified by RT-PCR. P19 cells grew and reached confluence within 48 h after the plating. Metformin suppressed the proliferation of P19 cells in a concentration-dependent manner. Among several cytokines, gene expression of tumor necrosis factor (TNF)-beta increased by metformin in a concentration dependent manner. It was therefore suggested that metformin-induced suppression of the proliferation in P19 cells might involve the alteration of the gene expressions and secretions of TNF-beta.
