IL-18 does not modulate dectin-1 expression on human monocytes challenged with Paracoccidioides brasiliensis

IL-18 is a proinflammatory cytokine and its role in paracoccidioidomycosis, a deep mycosis caused by Paracoccidioides brasiliensis (Pb) is not fully understood. Recently, we demonstrated that this cytokine, by modulating TLR4 and MR expression, play an important role on human monocytes response to P. brasiliensis. The aim of this study was first to evaluate the involvement of dectin-1 on IL-18, TNF-α and IL-10 production by human monocytes in response to P. brasiliensis. Moreover, we asked whether IL-18 can modulate the production of these cytokines by altering the expression of this receptor. Monocytes were stimulated with IL-18 and treated with specific neutralizing antibody anti-dectin-1 before challenge with more virulent (Pb18) and less virulent strain (Pb 265) of the fungus . After, dectin-1 expression was analyzed by flow cytometry and IL-18, IL-10 and TNF-α production by ELISA. Results showed that higher levels of IL-18 and IL-10 were detected after challenge with Pb 18, in comparison to Pb 265. By other hand, Pb 265 strain induced higher levels of TNF-α. However, only TNF-α induced by the strains were decreased after blocking dectin-1 receptor, showing the involvement of this receptor only in the production of this cytokine. Moreover, cells preincubation with IL-18 did not affect cytokines production by monocytes in response to P. brasiliensis, as well as dectin-1 expression. We can hypothesize that at least in relation to dectin-1 receptor expression, lack of modulation by IL-18 could contribute to susceptibility of the response against the fungus.
